Effets d'un mélange de pesticides sur les hémocytes d'huître creuse, Crassostrea gigas. ArchiMer
Gagnaire, Beatrice; Geret, Florence; Haure, Joel; Martin, Jean-louis; Burgeot, Thierry; Thomas-guyon, Hélène; Renault, Tristan.
Shellfish industry is often realised in estuarine areas, subjected to pollutions due to agricultural activities. The harmful effects of pesticides on animals inhabiting these estuarine zones are poorly documented. Bivalve molluscs represent an interesting model because they are sedentary and they filter water intensively. Effects of pesticides on haemocytes functions were tested in Pacific oyster, Crassostrea gigas and analysed by flow cytometry. A combination of pesticide at environmental concentrations was tested in vivo. Animals used corresponded to two genetically selected populations produced in hatchery.

Barranger, Audrey.
The aims of this PhD were to determine (1) the effects of the herbicide diuron on the genome of the oyster, Crassostrea gigas, (2) the possible transmission to the offspring of damaged DNA and (3) its consequences on oyster physiology. The genotoxicity of diuron and the vertical transmission of DNA damage were highlighted after parental exposure to environmental concentrations of diuron during the gametogenesis. Primary DNA lesions were detected in both somatic cells and germ cells of exposed genitors. Detection of the oxidized base 8-oxodGuo in gonads shows that exposure to diuron led to an oxidative stress which likely explains the observed DNA strand breaks in spermatozoa. The transmission of these DNA damage was confirmed by fluorescent in situ...

Etude des effets de polluants sur les paramètres hémocytaires de l'huître creuse, Crassostrea gigas - Interactions entre environnement, mécanismes de défense et maladies infectieuses ArchiMer
Gagnaire, Beatrice.
Shellfish industry is mostly realized in estuary zones, which are subjected to pollutions due to anthropic activities. The harmful effects of pollutants on animals inhabiting these estuarine zones are poorly known. Among these animals, Pacific oyster, Crassostrea gigas, may represent a model because they are sedentary and they filter water intensively. Among all physiological functions possibly disturbed by pollutants, defence mechanisms are poorly studied in bivalves. Moreover, animals presenting impaired defence mechanisms may be more sensitive to infectious diseases. In this context, effects of pollutants on hemocyte functions and on sensitiveness to diseases were tested in C. gigas. After adjusting the protocols for the monitoring of hemocyte...

In the last decades, shellfish industry represents an important economic activity in France and particularly in Charente-Maritime. Marennes-Oleron basin is the first French area of oyster production, especially Pacific oyster, Crassostrea gigas. But this area is also subjected to many pollution due to anthropic activities. Industrial wastes are sources of mercury pollution in the Charente river. The harmful effects of such contaminants on animals inhabiting these estuarine zones are poorly known, although many studies begin to be developed. Most of these studies use bivalve molluscs as biological indicators : these animals can bioaccumulate pollutants within their tissues by filtration. Moreover, they are sedentary so they cannot escape in the case of a...

Eutrophication in a tropical pond: Understanding the bacterioplankton and phytoplankton dynamics during a vibriosis outbreak using flow cytometric analyses ArchiMer
Lucas, Ronan; Courties, C.; Herbland, Alain; Goulletquer, Philippe; Marteau, Anne-laure; Lemonnier, Hugues.
In tropical shrimp ponds, the increasing of feed input, concomitantly with the stocking shrimp biomass, induces an eutrophication of the ecosystem. Although difficult to maintain, its stability is required to guarantee the success of the culture. A 110-day period of phytoplankton and bacterioplankton stock and dynamics in an earthen pond (1.2 ha area, 1 m depth) was monitored using flow cytometry to provide baseline information on community characteristics and ecosystem instability. Seven autotrophic cell types were identified over the whole sampling period. Prokaryotic cells included Synechococcus sp., a group named UNK which presented an atypical new flow cytometric signature and picoeukaryotes (PEUK). Nanophytoplankton cells were represented by 4...

Evaluation of annexin V and Calcein-AM as markers of mononuclear cell apoptosis during human immunodeficiency virus infection BJID
Palma,Priscila F.R.; Baggio,Giovana L.; Spada,Celso; Silva,Renata da; Ferreira,Silvia Inês A.C.P.; Treitinger,Aricio.
Evaluation of apoptosis by flow cytometry is generally accomplished by methods that use annexin V-FITC as vital dye, which access phosphatidylserine exposed on the external membrane at the beginning of this process. In addition, the concomitant use of propidium iodide makes possible to verify the characteristic nuclear alterations in the late stages of apoptosis, as a consequence of the increase in membrane permeability. On the other hand, the use of calcein-AM in association with ethidium homodimer (EthD-1) allows the evaluation of cell apoptosis through detection of esterase activity and cellular membrane physical and chemical alterations. The aim of this study was to compare the sensibility of calcein-AM and EthD-1 with annexin V-FITC and propidium...

Evaluation of urinalysis parameters to predict urinary-tract infection BJID
Santos,Juliana Conrad dos; Weber,Liliana Portal; Perez,Leandro Reus Rodrigues.
We evaluated the performance of automated-flow cytometry, urinalysis dipsticks and microscopic urine sediment analysis as predictors of urinary tract infection. Urine cultures were used as a reference method for comparison. Six-hundred-seventy-five urine samples from hospitalized and not hospitalized patients attended at Hospital Mãe de Deus, Porto Alegre, in south Brazil, were included in the study. Among the individual measures analyzed, intense bacteriuria in the microscopic analysis of urinary sediment gave an accuracy of 92.9%. A combination between intense bacteriuria (microscopic analysis) and >20 leukocytes per µL of urine (flow cytometry) gave a higher accuracy (97.3%). We conclude that though it is laborious, microscopic urinalysis is a good...

Expression of class 5 antigens by meningococcal strains obtained from patients in Brazil and evaluation of two new monoclonal antibodies BJID
De Gaspari,Elizabeth N.; Zollinger,Wendell D..
Determining the profile of antigen expression among meningococci is important for epidemiologic surveillance and vaccine development. To this end, two new mouse monoclonal antibodies (MAbs) have been derived against Neisseria meningitidis proteins (class 5). The MAbs were reactive against outer membrane antigens and were bactericidal. Selected anti-class 5 MAbs [(5.1)-3E6-2; (5.3)-3BH4-C7; (5.4)-1BG11-C7; (5.5)-3DH-F5G9 also 5F1F4-T3(5.c)], and the two new monoclonal antibodies C14F10Br2 (5.8) and 7F11B5Br3 (5.9), were then tested against different meningococcal strains, (63 strains of serogroup A, 60 strains of serogroup C (from 1972 to 1974); and 136 strains of serogroup B (from 1992) meningococci). Our results demonstrated that the expression of class 5...

Flow cytometric assessment of morphology, viability, and production of reactive oxygen species of Crassostrea gigas oocytes. Application to Toxic dinoflagellate (Alexandrium minutum) exposure ArchiMer
Le Goic, Nelly; Hegaret, Helene; Boulais, Myrina; Beguel, Jean-philippe; Lambert, Christophe; Fabioux, Caroline; Soudant, Philippe.
The Pacific oyster Crassostrea gigas accounts for a large part of shellfish aquaculture production worldwide. Aspects of morphological and functional characteristics of oyster oocytes remain poorly documented, and traditional techniques, such as microscopic observations of shape or fertilization rate, are time and space consuming. The purpose of this study was to assess for the first time viability and reactive oxygen species (ROS) production of Pacific oyster oocytes using flow cytometry (FCM) and to apply this method to determine oocyte responses to in vitro exposure to the toxic dinoflagellate Alexandrium minutum. A culture of A. minutum caused a significant increase in oocyte ROS production, which gradually increased with the age of the culture, but...

Flow cytometry protocol to evaluate ionizing radiation effects on P-glycoprotein activity BABT
Santos,Neyliane Gonçalves dos; Amaral,Ademir; Cavalcanti,Mariana Brayner; Neves,Maria Amélia Batista; Machado,Cíntia Gonsalves de Faria.
The aim of this work was to establish a protocol to evaluate ionizing radiation effects on P-glycoprotein (P-gp) activity. For this, human peripheral blood samples were irradiated in vitro with different doses and P-gp activity was analyzed for CD4 and CD8 T lymphocytes through rhodamine123-efflux assay by flow cytometry. By simultaneous employment of percentage and mean fluorescence index parameters, subject-by-subject analysis pointed out changes in P-gp activity for some individuals and irradiated samples. Based on this work, the proposed protocol was considered adequate for evaluating P-gp activity on cells after radioactive stress. Besides, this research suggests that P-gp activity could be an important factor to define patient-specific protocols in...

Flow cytometry to measure impact of temperature and salinity on the survival of Bonamia ostreae, parasite infecting flat oyster Ostrea edulis, in seawater ArchiMer
Arzul, Isabelle; Bond, Celine; Gagnaire, Beatrice; Morga, Benjamin; Chollet, Bruno; Ferrand, Sylvie; Robert, Maeva; Renault, Tristan.
Bonamiosis due to the intrahaemocytic protistan parasite Bonamia ostreae is a European endemic disease affecting the flat oyster, Ostrea edulis. After its first description in June 1979ı in Brittany, the parasite rapidly spread to all French oyster farming areas and in other European countries through transfers of live molluscs. The parasite has been described in different ecosystems from estuaries to open sea and no clear correlations could he demonstrated between the disease development and environmental parameters like temperature or salinity. The infection can he directly transmitted by cohabitation between infected and non infected oysters. Nevertheless, the parasite life cycle, including its survival outside the host, is not completely...

Genetic diversity in a world germplasm collection of tall fescue Genet. Mol. Biol.
Cuyeu,Romina; Rosso,Beatriz; Pagano,Elba; Soto,Gabriela; Fox,Romina; Ayub,Nicolás Daniel.
Festuca arundinacea Schreb., commonly known as tall fescue, is a major forage crop in temperate regions. Recently, a molecular analysis of different accessions of a world germplasm collection of tall fescue has demonstrated that it contains different species from the genus Festuca and allowed their rapid classification into the three major morphotypes (Continental, Mediterranean and Rhizomatous). In this study, we explored the genetic diversity of 161 accessions of Festuca species from 29 countries, including 28 accessions of INTA (Argentina), by analyzing 15 polymorphic SSR markers by capillary electrophoresis. These molecular markers allowed us to detect a total of 214 alleles. The number of alleles per locus varied between 5 and 24, and the values of...

Genome size and ploidy variations in pearl millet (Pennisetum glaucum) and napier grass (Pennisetum purpureum) genotypes Acta Agron. (Palmira)
Animasaun,David Adedayo; Morakinyo,Joseph Akintade; Mustapha,Oba Toyin; Krishnamurthy,Rama.
Abstract Pennisetum glaucum (Pearl millet) and P. purpureum (Napier grass) are economically important members of the genus Pennisetum . The knowledge of variability in chromosome number, size and genomic content of the species could provide clues on the mechanisms responsible for decrease or increase in genomic content in the evolutionary pathway. In the present study, twenty nine genotypes consisting of 24 pearl millet and 5 Napier grass were assessed for inter- and intra-variations in chromosome number, ploidy status and genome content. Conventional cytogenetic was used for chromosome counting and flow cytometry technique for assessing genomic contents of the genotypes. Pearl millet genotypes were diploid (2n = 14) while Napier grass were mainly...

Genomic abnormalities affecting mussels (Mytilus edulis-galloprovincialis) in France are related to ongoing neoplastic processes, evidenced by dual flow cytometry and cell monolayer analyses ArchiMer
Benabdelmouna, Abdellah; Saunier, Alice; Ledu, Christophe; Travers, Marie-agnes; Morga, Benjamin.
In the context of the abnormal mass mortality of mussels in France since 2014, Flow CytoMetry (FCM) was used in 2015 and 2016 to study the DNA content and cell cycle characteristics of hemic circulating cells collected from 2000 mussels. The mussels were sampled from 12 wild and cultivated blue mussels stocks distributed along the French Atlantic coast from the south Brittany to Pertuis Charentais areas. During these surveys, various genetic abnormalities were frequently detected, and ploidy characteristics revealed contrasting profiles that corresponded to respective contrasting sanitary status, i.e. healthy mussels with high cytogenetic quality (HCQ) versus diseased mussels with low cytogenetic quality (LCQ). In the present work, FCM and hemocytology...

Goat umbilical cord cells are permissive to small ruminant lentivirus infection in vitro BJM
Martins,Gabrielle R.; Marinho,Rebeca C.; Bezerra Junior,Rosivaldo Q.; Alves,Antoniel de O.; Câmara,Lilia M.C.; Albuquerque-Pinto,Luiz C.; Teixeira,Maria F. da S..
Abstract Small ruminant lentiviruses isolated from peripheral blood leukocytes and target organs can be propagated in vitro in fibroblasts derived from goat synovial membrane cells. These cells are obtained from tissues collected from embryos or fetuses and are necessary for the establishment of the fibroblast primary culture. A new alternative type of host cells, derived from goat umbilical cord, was isolated and characterized phenotypically with its main purpose being to obtain cell monolayers that could be used for the diagnosis and isolation of small ruminant lentiviruses in cell culture. To accomplish this goal, cells were isolated from umbilical cords; characterized phenotypically by flow cytometry analysis; differentiate into osteogenic,...

Hematological and immunological development from birth to six months of age in Holstein calves Arq. Bras. Med. Vet. Zootec.
Baccili,C.C.; Oliveira,S.M.F.N. de; Costa,J.F.R; Massoco,C.O.; Pozzi,C.R.; Gomes,V..
ABSTRACT The hematological and immunological development of calves from birth to 6 months of age was performed by hemogram and cellular phenotype. Ten male Holstein calves were assessed in 13 moments: before colostrum intake (D0), every 2 days until the 10th day of life (D2 to D10), at the 15th day after birth (D15), and then monthly up to 6 months (D180). Calves presented hemoconcentration on day (D) 0 and showed a gradual decrease in hematimetric rates until D180. The inversion of the neutrophilic for lymphocytic profile was observed on D4. The percentage of CD3+ cells on D10 was higher than D30 up to D180. The number of CD4+ and CD8+ cells did not change between time points. The number of CD21+ lymphocytes was significantly higher at early time points...

Hemocyte characteristics in families of oysters, Crassostrea gigas, selected for differential survival during summer and reared in three sites ArchiMer
Lambert, Christophe; Soudant, Phillippe; Degremont, Lionel; Delaporte, Maryse; Moal, Jeanne; Boudry, Pierre; Jean, Frédéric; Huvet, Arnaud; Samain, Jean-francois.
High variability among individuals is often encountered when hemocyte characteristics are measured in bivalves. Such variability is suspected to result partly from genetic factors. In this study, hemocyte characteristics of six families of Crassostrea gigas were compared by flow cytometry at one sampling date in October 2001. These families were obtained from a nested, half-sibling cross design, and reared from July to October 2001 at three sites distributed along the French Atlantic coast from north to south: Baie des Veys (Normandy), Riviere d'Auray (Brittany) and Ronce (Marennes-oleron Basin, Poitou Charentes). Among the 15 measured hemocyte characteristics, production of reactive oxygen species (ROS) of untreated hemocytes (maintained in filtered...
